Effect of Recombinant Human Growth Hormone on the Proliferation of Human Leukemic Cell Lines


Abstract
As the number of long-term survivors of childhood leukemia increases, growth retardation has emerged as a significant complication. Treatment of these children with growth hormone has been suggested and sporadically implemented.
We, therefore studied the effect of recombinant human growth hormone(rhGH) on the proliferation of normal bone marrow cells and leukemic cell lines in vitro.
In order to investigate the effect of rhGH on normal bone marrow cells and several leukemic cell lines, the cells were cultured in medium with the concentration of rhGH 150, 200, 250ng/ml and without rhGH for 4 days. Leukemic cell lines, we used,

used, were REH(pre-pre B acute lymphocytic leukemia), MOLT-3 (T-acute lymphocytic leukemia), HL-60 (acute promyelocytic leukemia) and K562 (chronic myelocytic leukemia).
@ES The results were as follows;
@EN 1. At the concentration of rhGH 150ng/ml, there were signifcant proliferation of normal bone marrow cells and HL-60 cell lines compared with those of corresponding control groups.
2. At the concentration of rhGH 200ng/ml, there were significant proliferation of normal bone marrow cells and all of the leukemic cell lines except MOLT-3 compared with those of control groups.
3. At the concentration of rhGH 250ng/ml, there were significant proliferation of normal bone marrow cells and all of the leukemic cell lines compared with those of control groups.
4. Cultured colony index of the concentration of rhGH 150ng/ml, 200ng/ml, 250ng/ml showed a significant dose-dependent increment in normal bone marrow cells and all of the leukemic cell lines.
Our in vitro data suggest that circulating human growth hormone promote leukemic blast cell replication in vivo, and the supplemental adiministration of human growth hormone to leukemia patients in remission must be carefully monitored for early
relapse.
